In Jujutsu Kaisen, all living beings manifest a force called Curse Energy (呪力, Juryoku), which is based on negative emotions that naturally flow throughout the body. The average person can’t control this flow in their body. As a result, they lose their curse power every time. This resulted in the birth of the Damned (呪い, Noroi), spiritual beings whose great desire is to harm humanity. These curses take the form of fearsome monsters, including onryō (vengeful spirits), yūrei (ghosts), and yokai.

A Jujutsu (呪術師, Jujutsushi, literally "Cursed Technique Master" or "Wizard") practitioner is someone who has the ability to use the cursed power of his body. This helps to utilize this energy as needed, and reduces the release of such energies. High-level magic and curse users can tap into this power and use it to perform cursing techniques (呪術式, Jujutsushiki) that are often unique to the user or their family. An advanced form of the curse technique is Domain Expansion (領域展開, Ryoiki Tenkai), where the user creates pocket dimensions of varying sizes within the domain where all attacks are guaranteed to hit their target and increase in power.

Itadori Yuji is a high school student with extraordinary physical strength. One day at school, his friends untie an amulet from a rotting finger, which incurs a curse. Yuji swallows the finger to protect Jujutsu magician student Megumi Fushiguro - who collected it on her bed - but unwittingly becomes the host of a powerful ancient magician, Ryomen Sukuna. All magicians must exorcise the curse of Sukuna and Yuji. However, Gojo Satoru - Mei's teacher and the most powerful magician in the modern Jujutsu world - notices his ability to control his body and takes him to Tokyo Prefectural High School for training. Gojo postpones Yuji's execution until he eats all twenty of Sukuna's fingers so that the curse can be permanently lifted with his death.

Meanwhile, a group of clever cursed spirits plot an elaborate attack to reshape the world of Jujutsu as they see fit, starting with locking Gojo away because he is too powerful to kill and his presence maintains order in the Jujutsu world. The group's leader is the mysterious wizard and curse-user Suguru Geto, who was killed by Gojo a year ago. His team includes the sadistic Curse Mahito, Fire Curse Jogo, and Nature Curse Hanami. Gojo's first-year students - Yuji, Megumi, and Nobara Kugisaki - train to perform missions to exorcise low-level curses; during this time, Sukuan develops an interest in Megumi because of her power and potential. After Mahito kills his friend Junpei Yoshino, Yuji is heartbroken and experiences firsthand the harsh reality of the Jujutsu world he inhabits. During a tournament between the Tokyo and Kyoto Jujutsu schools, Mahito steals three "Death Paintings" - half human, half curse - from the Tokyo school and summons them in their full form. Yuji and Nobara kill two of them during a mission, leaving Jogo alone to survive.

Geto and his team begin their ultimate project on Halloween night in Shibuya. Gojo manages to kill Hanami, but is successfully sealed inside a special Geto artifact. It turns out that the curser is not actually Geto, but Geto's corpse, possessed by an ancient evil wizard called Kenjaku. After Gojo's death, the curse wreaks havoc, causing casualties and chaos. Yuji is defeated by Choso, but Choso sees a strange vision and discovers that he and Yuji are actually blood brothers, and Yuji's life is saved. Chogo force-feeds the unconscious Yugi with all ten of Sukuna's fingers at once, giving Sukuna enough power to temporarily control his body. Sukuna kills Jougo and battles with the shikigami that Kei cannot control, nearly destroying Shibuya in the process. After regaining consciousness, Yuji is heartbroken to witness Mahito kill his mentor and fellow Gojo colleague Nanami Kento, and severely injure Nohara. With the help of Kyoto student Fujido Aoi, he manages to defeat the real one, but Kenjaku absorbs the curse and awakens many new wizards he has cultivated over thousands of years across Japan, ushering in a new world of cursed spirits.

The Jujutsu high command then once again imposes summary execution on Yuji and appoints second-year Yuta Okkotsu as the executioner. Loyal to Gojo, Yuta helps Yuji fake his own death and joins forces with him to find Master Tengen - an immortal wizard who is half cursed and half human. Tengen reveals Kenjaku's plan to merge the former's consciousness with a Japanese human through the "Killing Game" - a royal battle between all awakened wizards to accumulate large amounts of cursed energy. Yuji and his allies search for a mysterious woman known as "Angel" who can free Gojo and help save Megumi's niece Tsumiki from participating in the Crying Game. Kenjaku captures Tengen, and after meeting Angel, Yuji and Megumi learn that Tsumiki's body has been possessed by the ancient wizard Yorozu. Sukuna takes possession of Yuji's body, ripping off a finger and forcing Megumi to swallow it, thus leaving Yuji's body and transforming into Megumi. By weakening Megumi's resolve, Sukuna's spirit will no longer fight Sukuna's possession, thus killing Yorozu, and also killing Tsumiki.

Main Diesel Unit Gojo. A few days later, Gojo battles Sukuna and nearly wins, but is killed, but not before his strength is greatly weakened. Kenjaku encounters Fumihiko Takaba, a wizard who effectively distracts Kenjaku, catching Yuta off guard and beheading him. Kenjaku makes Sukuna the master of the Crying Game before dying.
